eliciting th1 immune response using casein- (alpha s1) loaded dendritic cells
Authors
abstract
allergen-specific immunotherapy (ait) has been recently considered as an alternative approach to ameliorate the symptoms of allergen exposure and improvement the patients’ quality of life. dendritic cells (dc) in the forms of tolerogenic or th1-induced cells have been investigated in several studies as one of the promising approaches of ait in allergic diseases. the aim of this study was to evaluate the potency of casein-loaded dcs in eliciting the th1 immune responses in balb/c mice as a potential therapeutic approach in allergic condition. immature bone marrow-derived dcs were loaded with casein (protein or mrna) or green fluorescent protein (gfp) mrna. dcs were evaluated based on the expression of specific markers and production of proinflammatory cytokines. proliferation and cytokine production of lymph node lymphocytes and splenocytes were measured in dc-injected mice. expression of dc markers in all groups was significantly higher than immature dcs, but lower than lps-activated dcs. despite an increase in tnf-α and il-12, il-6 was decreased in casein-dc treatments. caseinloaded dcs could induce proliferation in lymphocytes and stimulate them to produce higher amounts of ifn-γ and in some extent il-10 and tgf-β, while they could not stimulate il-4 secretion. casein-loaded dcs could partially elicit the th1 responses; this would be a promising approach to use them as an allergic protective way for applying immune cell therapy in cow’s milk allergy.

full textevaluation of different listeria monocytogenes components on dendritic cells for eliciting th1 response
objective: dendritic cells (dcs) play a critical role in the beginning and in the course of immune responses. by observing. considering the defect in these cells in patients with tumor malignancy. in recent years there has been considerable interests in correction and use of these cells. in our prvious studies we showed that lysate of listeria monocytogenese can induce maturation of dendritic c...

Dendritic cells (DCs) induce pathogen-specific T cell responses. We comprehensively studied the effects of addition of maturation stimulus, fibroblasts (fibroblast conditioned medium), PHA activated T cells (T cell conditioned medium), and mixture of fibroblast & PHA activated T cells (FCM-TCCM) conditioned media on maturation of DCs. Monocytes were cultured with GM-CSF and IL-4 for five days. ...
full textIMMUNOBIOLOGY CD38 orchestrates migration, survival, and Th1 immune response of human mature dendritic cells
CD38, an ectoenzyme and a signaling receptor, is a novel marker of human mature monocyte-derived dendritic cells (MDDCs). The working hypothesis is that CD38 is not only a marker but also contributes to functions specifically gained by MDDCs with maturation. This was tested by assessing the role(s) of CD38 after signaling with agonistic anti-CD38 monoclonal antibodies or by blocking the interac...

full textDendritic cells in immune response induction.
The study of dendritic cells (DCs) has seen a rapid expansion in recent years, and their importance within the immune system is now widely recognized. Along with B lymphocytes and mononuclear phagocytes, DCs make up what are known as the professional antigen-presenting cells (APCs). These are cells which are capable of highly efficiently presenting antigens to the immune system in the context o...
